Det inducerbara anti-oxidantskyddet i hjärnan
Hjärnans celler är känsliga för oxidativ stress, ett tillstånd där halten fria radikaler överskrider anti-oxidantskyddet. Ett intakt skyddssystem som kan uppregleras (induceras) vid oxidativ stress är viktigt för att inte hjärnan skall skadas av reaktiva ämnen. Vi har studerat transkriptionsfaktorn Nrf2 som kan aktiveras av icke-toxiska nivåer av fria radikaler och ämnen i vissa grönsaker, t.ex. sulforafan i broccoli. Aktivering av Nrf2 leder till att ett 100-tals skyddsgener (över)uttrycks och hjärnan får därigenom ett starkare skydd mot oxidativ stress. Vi har visat att inflammatoriska ämnen kan leda till att Nrf2-funktionen underuttrycks i den vanligaste celltypen i hjärnan, astrocyten. En sådan nedreglering av Nrf2-systemet sker på människa i det minnesområde som skadas tidigt vid Alzheimers sjukdom, hippokampus. Vi tror att denna nedreglering i astrocyterna kan orsaka nervcellsdöd (astrocyten skyddar normalt nervcellen) och minnesbortfall i Alzheimer’s sjukdom. Vi har delvis identifierat de molekylära faktorer och signaleringen som reglerar Nrf2-systemet i odlade astrocyter. Aktivering av kinaserna p38 och GSK3beta nedreglerar medan aktivering av ERK, JNK och Akt uppreglerar Nrf2-systemet. Vidare aktiveras histondeacetylaser i astrogliaceller när de utsätts för inflammatoriska ämnen. Histondeacetylaser är enzymer som påverkar genuttrycket utan att förändra den genetiska koden, så kallad epigenetisk påverkan. I vår nuvarande forskning studerar vi hur Nrf2-systemet påverkas av stresshormon, vissa miljögifter, cytokiner och fysisk aktivitet. Vi är speciellt intresserade av epigenetiska faktor, som vi på goda grunder tror nedreglerar Nrf2 även i nervceller, mikrogliaceller och i hippokampus på individer som lider av Alzheimer’s sjukdom.
